Entwicklung und Validierung stabilitätsindizierender Prüfverfahren
Im Rahmen der Entwicklung von Prüfverfahren in der Reinheitsanalytik muss belegt werden, dass die Prüfverfahren "stabilitätsindizierend" sind. Damit ist gemeint, dass alle (potenziellen) Abbauprodukte als Verunreinigungen selektiv erfasst werden können. Um dies belegen zu können, muss im Rahmen von Studien zum forcierten Abbau, die in Europa auch "Stressstudien" genannt werden, der Wirkstoff unter unterschiedlichen Bedingungen abgebaut werden und so die potenziellen Abbauprodukte erzeugt werden. Dabei ist der Beleg der Massenbilanz ein zentrales Kriterium dafür, ob von dem entwickelten Prüfverfahren alle Abbauprodukte detektiert werden. Es muss dann gezeigt werden, dass das entwickelte Trennverfahren eine ausreichend selektive und empfindliche Bestimmung dieser (potenziellen) Abbauprodukte neben anderen Verunreinigungen ermöglicht. Falls im Rahmen von Stabilitätsstudien Gehalte von Abbauprodukten über die Qualifizierungsgrenze ansteigen, müssen diese qualifiziert werden.
